mRNA Quantitation

Dear Netters:

I am carrying out an analysis on winter flounder (Pleuronectus americanus) 
pituitary mRNA.  I am doing this by the use of slot blots dot blots and 
northern blots.  However, my quantitation doesn't seem to be all that 
consistent.  I am using a cDNA probe and random primers.  I was wondering 
if anyone would be able to suggest a suitable protocol for me to use for 
solution hybridizations using my cDNA probe.
