PCR thermal cycler operation

This question is directed at people whose thermal cyclers 
offer a choice between an in-tube temperature sensor and one 
that senses the temperature of the heating block.

Since the responses of different machines differ, I will be 
particularly interested in replies from those who use the 
Intelligent Heating Block from Cambio Ltd., Cambridge, 
England, but other experiences are also welcome.

The question is whether I can expect to move from using the 
in-tube sensor (with volume always adjusted to match sample) 
to the block sensor without any substantial effect on 
amplification results.

As usual, I will summarise to the net if justified by 
rsponses received.
