Although many feel that residual PEG is a big deal in preparing single
stranded sequencing templates, Pheiffer and Zimmerman (NAR 11(2)p.7853
1983) report that 95% of the PEG 6000 from a 15% solution is removed
from the aqueous phase by a single extraction with 3 vol. chloroform
at room temp. It seems then quite unlikely that PEG would contaminate
sequencing templates prepared with traditional organic extractions.
Perhaps this belongs in Ed's urban myth collection. Thanks for the
