We always quantitate DNA using Hoechts dye and the Hoefer TKO 100 fluorometer. Our results are much more reliable than 260/280 results. You must use dust free conditions, handle the cuvette with gloves, and practice, but once you've learned how to do it, the results are reproducible and reliable.
