TA Cloning

Dear Rick (and other netters),

A number of methods have been published for cloning PCR products and creating
ones own vectors to do so.  I have made one too, based on Bluescript II SK
plus.  It is published in: PCR, Methods and Applications Vol 2 pp81-82.  If you
like the sound of it you can certainly e-mail me and I'll send you some.  

Basically it is a vector that contains sites for restriction enzymes that when
cut leave either a 1, 2, or 3 base T-overhang as desired.  Personally I use the
3 base overhang and add a couple of extra As to the PCR product with Terminal
Transferase.  It works very well for me.
