antisense oligos

I want to use antisense oligonucleotides to inhibit the synthesis of my
favourite protein in cell culture. A previous reference used all
phosphorothionate linkages, but this makes the whole procedure very
expensive. I have heard that using thiol substitutions at all points is
not really necessary, but that using substitutions at the terminal
couplings only will prevent exonuclease digestion, which is the major
degredative pathway. Is this true? Has anybody done this and shown that the
different oligos are equipotent or nearly so?

Any advice gratefully received,

michael duggan
