Potentially yes, it may acylate. But no one has told the acetic acid or the protein yet so whilst they exist in blissful ignorance we still get good sequence. We run the sds gels with thioglycolate as well. I haven't had any problems the times I forget to add the thioglycolate. This set up works in our hands quite well.. ..d
