If your PCR reaction is not problematic (you have long enough primers and small
 enough fragment) you can PCR the cells directly. In our lab we grom E. Coli ce
lls over-night in LB, then take 10 microliter directly for PCR. I read in BioTe
chniques (sorry but I don't have the exact referance) that Microwaving the cell
s should help. Didn't try that yet, but it sounds reasonable.
