Dear netters I have been trying to clone an oligonucleotide into pUC 19 derivative plasmid pWM 529 using Mandecki`s single-stranded bridge-repair protocol. I have followed the protocol very carefully but still have very little recombination if at all. Does anyone out ther have some advice?
