Preferential insert orientation

This is a very common phenomenon with cloning.  It has happened to me
numerous times as well as with others in the lab.  Really the best you
can do besides work yourself into the gound over it, is to chalk it up to
"Mol. Bio voodoo".  I know its bad to admit but there's still quite a bit
that is not understood about the molecular details of cloning........Good
