One other thing about cloning PCR fragments with T vectors just came to me. If you use polymerases other than TaqPol (Vent, etc) you don't get as many A overhangs on your PCR product. We have demonstrated this indirectly by getting fewer clones from Vent amplified reactions. Everbody may already know this, but I thought I'd throw it out there! Bruce
