There is an excellent reference called "Molecular Cloning: A Laboratory
Manual" by Sambrook, Fritsch, and Maniatis. Around here we just call
it the "Maniatis Manual". It is filled with time tested methods that 
will easily replace the many expensive kits that people have become
convinced are the standard in today's molecular biology laboratory.
G-50 column preparation is discussed in Appendix E pg.37.
