Expressed protein extraction from cos cells?

Hello All,

  What is the best way to extract expressed proteins from cos cells? 
Should one use a polytron? sonication?  What buffers should be used?  If
the protein is similar to cytochrome P-450s and has large hydrophobic 
surfaces, are different buffers called for?
  I will be happy to post a summary of any responses.
