this idea is not that unique. check out Gene v77 (1989) pp51-59. i have tried
their method and it worked well for me. one difference is that they do two
separate reactions to amplify each end, then do a reaction using the two
ends to prime each other and including the outside primers. did i explain this
clearly? anyways, pick up this paper. i tried to do all reactions in one tube
but got such a gamish of stuff, it is best to stick to 3 reactions, although
i did not bother to clean up the products of the first pair of reactions.
