ligations in LMP agarose

You should remove most of the LM agarose by freezing the slice and then
spining out the debris. This gives only a small (four-fold) reduction in
ligation-transformation efficiency as compared to samples without any
agarose (as reported to me by Mike Coady <coady at>). Maniatis
describes a 100-fold reduction associated with ligation in melted agarose.

See the FAQ or search the archive at by gopher on
"consistent and subcloning" for more details.

Sorry for the broken record effect.
