Using glycerol to enhance PCR amplification.

	A few days ago, there was a request from someone who would like to
know the real effect of glycerol in enhancing pcr amplification. I may
suggest him to read "Amplification", Sep 1990 Issue 5 published by the
Perkin-Elmer Corporation. In brief, you can add 5-20% of glycerol into
your PCR reaction mixture. It will effectively increase the amount of your
amplified product from G/C-rich DNA template. I always add 10% of glycerol
in order to get 950 bp amplicon from G/C-rich Herpes simplex virus genome
(template). I guest that glycerol might help PCR reaction mixture to
accumulate enough heat to denature G/C-rich double stranded DNA template 
and do also stabilize Taq DNA Pol at the same time. Sorry in advance if I 
am wrong.
