I'm looking for references or protocols to use AP or HRP conjugated to
streptavidin or avidin for the rapid detection of biotinylated DNA. My
goal is to develop a rapid procedure for determining whether a PCR product
has been produced. The procedure is to be used by folks that do not
normally run gels and who simply want a yes/no answer.
The design is:
1. Do PCR with biotinylated DNA.
2. Separate possible PCR product from primers by ultrafiltration,
chromatography, precipitation, etc.
3. Add AP conjugated to streptavidin and then wash.
4. Add AP substrate and look for color development.
