Isolation of Nuclei

Does anyone have any experience in isolating nuclei from human leucoytes
(or any cell line)?  Since we are interested in doing in situ 
hybridization, we would like to avoid including any detergent during the
isolation process.  Detergent will destroy nuclear structure.  Any 
protocol will be greatly appreciated.  Thank you.
