I have recently seen a protocol for producing ssDNA templates for sequencing by performing a PCR reaction on 2-10% of a PCR product using only one primer. I have successfully amplified a ssDNA template this way but have not yet sequenced it. Has anyone out there been successful with this technique, if so is there anything I should know before starting?
