Protein expression

WE are trying to express some ECM receptor cDNAs in E coli and are having
no luck. The cDNA being expressed contains no signal peptide, or transmembrane
domains, and is not being expressed as a fusion protein. Upon induction we
get nothing. The control plasmid works well under the same circumstances. The
insert is in the right frame and orientation. Does anyone have any suggestions
as to what I can do ? Are there any problems associated with expressing extra-
cellular molecules in bacteria. A quick review of the literature reveals that
most antibodies are raised against synthetic peptides. Is this significant ?
Any help appreciated as with no expression it is hard to decide what to change.
Thanks
Mungo
