AMPLIFY A LIBRARY

Hi.

I would like to amplify a plasmid cDNA library in E. coli.
I have about 10 micrograms to start with.

I would like to do the transformation and then grow the transformed bugs 
in culture, (NOT on plates).

One Question I have is: 

After heat shocking the transformations,
exactly how long (and in what volume) is best to grow the culture before 
working it up for Cesium Prep?

Any general suggestions for amplification are also welcome!
