In designing degenerate oligos from an amino acid sequence is there:
1. A temperature I should shoot for
2. A length I should shoot for
3. A difference between using GATC and I in the wobble position if all variations are desired
Please write me with any advice
I am using codon preference for the organism from which I will be amplifying.
