Dear Fimme, A couple of good references for NTCB chemistry are Degani and
Patchornik 1974. Biochemistry 13:1-11 and Jacobson et al. 1973. J. Biol. Chem.
248: 6583-6591. Its clear that use of this reagent has several possible
outcomes, dependent most on the initial reaction conditions (ie. pH). I've used
NTCB in peptide mapping experiments, and although the results have been
interesting, they could not be interpreted simply as cleavage at cysteine
residues. Liao and Wadano 1979 ( J. Biol. Chem. 254: 9602-9607) have reported
that NTCB also modifies and cleaves at serine and threonine residues.
