Please teach me CAT turnover rate

Tue Jul 26 06:18:09 EST 1994

Dear Netters

Does anybody know the turnover rate of chloramphenicol
acetyltransferase (CAT) in mammalian cells ?

We observed mRNA level of an enzyme decreases after addition of
forskolin in Northern blot analysis. We plan to transfect the promoter
CAT plasmid of the enzyme to the cell and to measure CAT activity after
addition of forskolin at 24-36 h after transfection.

In this case, can I expect that the CAT activity will decrease
immediately, if the transcription from CAT plasmid and the mRNA level
of CAT decrease. In this case, the turnover of CAT protein is important.
I appreciate it if you teach me.

Sincerely yours,
