Hi John,

We have used PCR mutagenesis quite successfully, but we do have a good 
selection/screening protocol to determine the mutants we want.
There is no single protocol, since the protocol will depend on the size of 
your PCR product and the number of mutants per product required.

There is a good review of PCR conditions and fidelity by Eckert and Kunkel (pg 
225-244)  in the book:

PCR - A Practical Approach

Edited by McPherson, Quirke and Taylor
