Buffering Phenol

A method for buffering phenol that always worked for us is described by
the Life Sciences -Gibco-BRL "Ultra Pure" laboratory handbook you can
get for free from the company (It has many protocols for routine
molecular biology reagent preparation).
1/ Melt phenol or start from water saturated phenol (either d=should
work)
2/ Mix with an equal volume of 0.5 M Tris-HCl (ph 8.0)
3/ Aspirate upper acqueous phase
4/ Add equal volume of 0.1 M Tris-HCl (pH 8.0). Repeat step 2
5/ Check pH with pH paper. Repeat steps 3 and 4 until equilibrated to
desired pH Two-three repetition usually yield pH 8.0
