The trick for directly PCRing lambda clones is to pick the plaque into water, not SM. All that Mg in the SM (and other phage storage media) *really* mess up the PCR. We pick a plaque to 100 ul H20 and then use 10 ul for PCR. Good Luck! Carlisle
