I just got the following results on a standard Northern Blot.
There was a slow-migrating smear whose intensity decreased
down the blot. Also, there was very faint hybridization with
the 18 and 28S bands. This implies nonspecific reaction of
the probe with the message, but it does not explain why the
slower smear was so dark. This pattern was seen with the
total RNA of only one of two cell types loaded onto the gel.
18 and 28S bands looked normal. 10ug/lane. Possible bands
were witnessed within the smear. Any idea what caused the
smear or what to do about it?
