In my experience Pfu gives better yields than Vent.  Both are more
finicky than Taq, the buffer sometimes needs to be optimized
(Stratagene supplies 2), and better luck can sometimes be had with the
addition of 10% DMSO or TMAC. I once had a problem with Pfu (or was it
Vent....) chewing the ends of my DNA, but that was solved by running
the cycler more promptly.
