help method for protein estimation

Hi friends,
The affi gel from Biorad according to the manufacturers instructions  the
time and efficiency of coupling of a ligand to affi gelis unstable in
aqueoous solutions.The alternmate way is to dissolve the protein ( ligand)
in an organic solvent in order to increase the efficiency of coupling . My
protein dissolves in formamide.
Is it possible to estimate the amount of protein in formamide by any other
method than measuring O.D at 280nm. And also can somebody tell how i can
get the back the protein from organic phase to the aqueous solution ?
