Loss of plasmid in liquid culture

Bill Studier!s paper on the pET vectors (Meth. Enzym. 185:60-89) points
out that using ampicillin for selection has inherent problems because
beta lactamase is secreted into the medium and can destroy all of the
ampicillin, and there can be enough in an overnight culture to wipe out
all of the ampicillin in the larger culture at the time of inoculation. 
The paper discusses various ways around the problem of plasmid
instability.  There might be something in there to help with your problem.
