We have used pfu in place of TAQ when we want fidelity. In our case we just decreased the annealing temperature around 5-10 degrees. Be sure to use the use the buffers supplied with the enzyme, not the TAQ buffer. TAQ buffer has sodium salts at 50 mM, which is detrimental to pfu. Good luck Neal Musto
