I've made a few constructs using recombinant/recombination PCR with one 
or two mutant primers, and it worked swimmingly the first time and poorly 
the second, trouble is its easy to set up in theory but the PCR may 
require a good deal of time consuming "tweaking" to produce enough of the 
mutated product to clone. When it works its damn fast and very neat, when 
it doesnt its a pain in the ass!
