One-Step Transformations

Hi Netters!

I have been trying to use the one-step transformation method (Chung etal.,
1989) to isolate constructs of 7.5-10.5kB (pBR322, HB101).  Although my
cells have taken vector without insert consistently well, I have had very
low efficiencies obtaining vectors with inserts as described.  These
constructs have since been isolated using electroporation.  Is there a
size limitation with this method in anyone's experience or is this just a
phenomenon based on Voodoo?
