Luciferase detection

Hallo everybody,

I transfected HeLa cells with a construct having the Luciferase gene as
a reporter gene. I would like to detect the positive clones directly in
the culture dish by adding the substrata (D-Luciferin etc..).
Has anyone done it ?
Does this procedure kill the cells ?
Can you detect the positive cells in situ ?
