Have a look at Maniatis or Current Protocols.

which is not covered by a book (ugh!), search the old bionet messages by

The one which gives you the most bang for the buck :-)

Generally, reading Maniatis (or something similar) and searching the 
archives would make many questions in this forum unnecessary.
