In response to Joanne Tornow:
I have used the method outlined in Kadonaga and Tjian (1986) PNAS 83,
5889-5893. It gives a complete protocol from start to finish including
activation of the resin (which can be skipped if using an already
activated resin), coupling of DNA to the resin, and the column itself.
It worked considerably better when I used a partially purified protein
fraction.
If you want more information, I will gladly relay my experiences with
the affinity column.
