isolation of nuclei from lymphocytes

I was wondering if anyone out there has a more efficient way of isolating
nuclei (for DNase 1 digestion) from lymphocytes (thymus, lymph nodes and
spleen). I have been using NP-40 overnight at 4 degrees C while the cells
are encased in low melting point agarose but I have gotten inconsisent
results between spleen and lymph node cells. Any suggestions?
