re: PCR from intact bacterial cells

Hello everyone:
I have done PCR from plasmid inserts by wetting a 
toothpick in a overnight E. coli culture or using 
the toothpick to touch a colony. The toohtpick is 
then used to shake a 10 ul reation. No treatment 
seemed necessary, perhaps the first 2 min at 95
degrees were enough to break the cells. 
It works quite well.
