In continuing Duncan's suggestion that heat induction was inadequate in the 500
ml culture I would like to add that adding hot media (just out of the 
autoclave you can calculate the appropriate volume quite easily) you can raise 
the temperature very quickly.  I have always done this for all my heat 
inductions and have been pleased with the results.
