direct PCR from bacterial colonies.

I normally pick the colonies with toothpick, suspend the cells in 50ul 
of water, vortex the tube, heat the tube at 99oC for 5 min and spin down 
the cells at 14,000rpm for 1 min.  I use 10ul of the supernatant for 
50ul PCR reaction. It works everytime.
