DNA "refusing" to be run in agarose gel

I have had this same problem.  I solved it by simply shaking (mixing) my
gel loading buffer.  The gel loading buffer has glycerol in it which makes
your sample stay in the well.  For some odd reason, the water and glycerol
in my gel loading buffer were not mixed well.  Upon shaking up my buffer,
the problem was solved.

Another thing you could try is to increase the amount of gel loading
buffer you have added to the DNA sample.  Hope this helps.
