Does anyone have a reproducible method to transiently transfect purified peripheral blood monocytes ? We have tried lipofection, DEAE-Dextran, calcium phosphate and electroporation but the levels of reporter gene expression are too low even with a very strong promoter. Any suggestions will be gratefully received. Thanks. John S.
