SEQUENCING Large Plasmids

When faced with this problem, I decide to add an F1 origin and generate
11 nt single-stranded circular plasmids for sequencing. Pharmacia's 
PucF1 vector provides a phage origin within a large polylinker.

Even large single stranded circles required gel purification 
after heat-SDS liberation of the DNA for clear sequencing. Contact
