I don't have much experience with S17-1, but I was able to obtain up to
10^7 cfu/ug S17-1/lambda pir using both Hanahan procedure and procedure
described in Chung, Miller Meth. Enzymol. v 218 p.621. I stick to the
latter one 'cos it simple and quite reproducible.
