DNA/RNA ratios as indicator of physiological activity

Seasons Greetings!

I have a few question concerning DNA/RNA ratios (as used as an indicator of
physiological activity).  I know that small quantities of DNA can be
quantified using the Hoechst Compound 33258 and a fluorometer.  How does one
quantify RNA to a similar level of sensitivity?  Can Acridine orange be
used?  Are standard extraction (CTAB) and ethanol precipitations sufficient
for this analysis.  Any help would be appreciated.  Thank you.
