I am trying to clone a gene into a plasmid using a unique PacI site. PacI leaves an AT overhange and I suspect this could be causing my problems. Any suggestions on what ligation conditions to use (ie ligation at 10C) would be greatly appreciated!! Thank you, Andrew Little
