Does anyone out there have a particular level of formaldehyde
to use in RNA denaturing agarose gels for northern hybridization analysis?
I've heard some different ideas, but no real reasons why.
My current recipe for a minigel is:
0.65g agarose
4.25ml 10x MOPS/EDTA
34.85ml water
cool to 50C
4.4ml stock (37%) formaldehyde
This yields a 1.5% gel with about 3.75% formaldehyde. Some suggest
more, some less. Any ideas?
