One of the problems with this kind of method is the amount of secondary
antibody. I use ECL. With one of the monoclonals, I have to use a dilution
1/20,000 for the anti-mouse-peroxidase; another way several proteins react.
Thus, one of the possibilities is that you determine which is the lowest
concentrations of secondary antibody that give you a response, and the same
for the primary antibody.
