Yeast Mito Regulation

Dear Yeast people,

        I am looking for two things,

1)  Is there a modern Mitochondrial Transcription Assay, The one I am
currenly using is quite old, and I am concerned about the efficiency.

2)  What is the general consensus on the activity of Nuclear-encoded
proteins BEFORE they are transported to the Mito?  I would Like to study
a Mito factor but I would rather express is from a plasmid and not have
to do Mito Preps. to get the functional protein.
